hi there... am a new one in field of cloning. i m trying to ligate my gene of interest in pET 23a vector but cudnt do that till date. how should i proceed???? thanx in advance.

It might help if you give some more details like what protocol you use etc... how do you try it?
If you don't know it, then ask it! Better to ask and look foolish to some then not ask and stay stupid.
