We are currently struggling with getting translation in fresh synaptoneurosomes. The question I have is about time intervals. First, to see the translation by WB - for example freshly produced camkII, how much time after stimulation is the best? Would it be rather three minutes or 20? I've searched the papers and they seem a bit inconclusive in our circumstances. Also, is there a possibility of translation in 4 degrees, in the control sample? It sits on ice for the same amount of time the other are incubated (up to 20 minutes).
