Hi,
I'm trying to CoIP an endogenous transcription factor from primary neuronal cells to identify interacting proteins by MS/MS and had the following concerns:

1. How much starting material should I use ? Currently I have about 10^7 cells per condition and my resource is limited since its dependent on the availability of animals.

2. This transcription factor is over-expressed in the mutant condition as determined by Western blot/qPCR, but I do not have any idea about the abundance of this protein.

Any suggestions will be greatly appreciated.
