Hi, I want to do fixation, dehydration, sectioning and H&E staining for my muscle samples. These samples were treated with 0.6 M NaCl, and this ionic strength will lead to the cell swelling. I also want to see this strucutral change in microscope at last.
In fixation step, I choose 4% paraformaldehyde in 10 mM PBS as the fixation solution. So for the reason above, should I raise the salt to 0.6 M also?
Thanks in advance.
