Macrophage dinamic in culture

Hello everyone!

I am new on working with macrophages culture (from mouse peritoneal cavity).. and I have some basic questions :lol::

1) I've seen that macrophages, in culture, start forming colonies or clusters.. and I canīt find any information in the literature about this.. does it always happen? why?

2) Does macrophages, once they adhere to the dish surface, keep gorwing?

3) Tripsyn is not enough to deatach them?

I haven't seen any clusters.
I do some primary macrophage culture, and they start forming multinucleate giant cells after a couple of days on the plate. I'm not sure what causes it. I've seen things such as acidic media or IL-4 can cause that to happen. I haven't figured out how to get them to last any longer, but I'm working on a few things right now. A colleague does the same thing I do, but in a different species and his cells last at least 5 days before fusing.

to 3) 10mM lidocaine+10mM EDTA in PBS.
