I have a serum antigen that I would like to eliminate from some mouse serum samples to test whether this factor mediates some effect of the serum on cultured cells. A neutralizing antibody against the human antigen is available, but this is only partially cross-reactive with its mouse antigen. Moreover, it costs a lot...
My question is therefore - could neutralization via incubation in an ELISA plate coated with a monoclonal capture antibody be considered an acceptable alternative? Has any of you ever seen such a method being utilized in previous publications? The effectiveness could of course be verified by comparing native and depleted serum via ELISA, so the question is whether this treatment would likely cause other changes to serum composition.
Of course I could use a protein A-sepharose depletion of the antigen, but that would remove serum Ig as well...
