has anyone tried to spin down nuclei and other cell debris AFTER adding laemmeli sample buffer (I forgot the spinning step - too many things going on)? I imagine SDS might crash out, but perhaps it could be added back. Thanks in advance!

the sds won't crash out. some of the debris will have solubilized so your sample will be more complex than it would have been if you had spun it first (it may also be more viscous due to the genomic dna from the nuclei).
