Hello everybody, i am having trouble with my expression. I want to express a beta glucosidase gene in pichia. I am using mgy buffer, after 18 h i solve the culture in mm medium. İ prepare all the mediums and components fresh. i got an absorbance once but when i add methanol 0.5%, the second day i can't see any activity. Most beta glucosidase assays were done by pichia and methanol induction. I dont think that methanol inhibits my enzyme, but i cant solve this problem if there is any clue for this please help me!! thanks and good luck...
