Any one can help me finding the best method of extracting mitochondria from human liver cells without damaging the membrane bound enzymes (outter and inner membrane bound).

Hi,

have a look at a mitochondria isolation kit, like from Qiagen. If you need further purification you can add up a gradient centrifugation. Additionally check the publications from H. Shaegger. His group is doing that frequently.
