Hello all!
I am triying to develop a Ni chromatography. I have the idea that you should use a pH of 8 to have your his-tag in positive charge (as the pKa of His is nearly 7, you have to take a lot of care with pH). I am using as buffer for diluting my seed, Borax 20 mM pH 10, i assume that once you are over the His pKa, the used pH  is irrelevant. Any of you have a clear idea about this issue?

Thank you in advance
