Is anyone aware of an equation to calculate or even a general idea of what the temperature (or Tm potential, due to the urea and formamide) is across a DGGE gel? I am attempting to figure out how much the temperature (melting potential) changes across a 35-65% gradient.

So for example, if a primer denatured in the 35% gradient of a DGGE gel, what would its approximate Tm be?
The same question for the 65% gradient on the DGGE.

Thanks in advance for any words of wisdom.

Liz
