How can I clone the upstream or downstraem of my gene??

I had spent alot of time on screening the uptream o my gene by colony hybridization.
However, the transform efficiency is very low..
My DNA fragments is about 3.8kb....
How can I clone the upstream or downstraem of my gene??
Please help me.

You will need to perform a process called inverse PCR.
