hi everyone,
I want to make Hepes-KOH buffer, but protocols always suggest me to filter the buffer not to autoclave it. Dose anyone know why?
thanks

heat (and even long-time storage) modifies HEPES; old HEPES gets light browny; I think of oxidations...

hi fren....
from my own experience...
better for u to sterile filter all buffers rather than autoclave due to changes of physical properties pH and conductivity sometimes and more serious for chemicals properties of the combine chemicals such as oxidation and so on...
