i am working with IgG1 with is suppose to have approx 16 disulfide bond (got this info from literature...) i would like to know how can i calculate no of moles —SS— groups in a protein ??? is there a therotical formula or way to do it..please let me know if so....thanks a lot

Uh, it is 16 times the number of moles of protein present. You can figure that out by knowing the molecular weight of the protein and the weight of your sample.

well the mol. wt of my protein is apprx 150 kDa and i am using 10 ug protein..so how do one go about and calculate the moles of -SS- bond????..is it therotical or experimental

moles of protein = weight/MW = 10 ug/150Kg = 6.7e-11 moles
moles of S-S bonds = 16 * moles of protein = 16 * 6.7e-11 = 1.07e-9 moles
moles of S-S bonds = 16 * moles of protein = 16 * 6.7e-11 = 1.07e-9 moles
