Hi all,
I transfected gaussia-luciferase gene to leukemic cell line cells. The cells were very bright after adding coelenterazine by both a luminometer and a xenogen imaging system in vitro. However, when I injected the cells s.c., i.p. or i.v. to mice, I couldn't detect any signal by xenogen imaging system.
I took picutres from 30sec to 5min after injection of coelenterazine.
One mg coelenterazine was disolved with 0.2ml ethanol and 1.8ml PBS. 0.2ml (100microgram) coelenterazine was injected to mice through tail vain.
Mouse's strain was NSG (NOD.Cg-Prkdcscid Il2rgtm1Wjl/SzJ JAX®)
Does anybody have any idea to solve this problem?
