Has anyone had any luck with combining different Pfu polymerases in quikchange style mutagenesis? My PI has heard of other groups successfully trying that but of course I don't find any information online about any attempts. We have used both the Quikchange II mutagenesis kit (successfully for a bunch of mutants) and then tried the quikchange lightning kit for some stubborn mutants we still can't get. With the Quikchange II kit we tried both Pfu Ultra which, worked on some mutants in the past, and Pfu Turbo, which I got to work on a bunch of new mutants.
