I need help in calculating the specific activity for complex IV(CIV). I have used a spectrophotometric method (550nm) to measure the oxidation of reduced cytochrome c, to obtain CIV activity. I have the rate of reaction (change in absorbance /time), and protein concentration in the sample (source of CIV).
.Can someone help me with the formula for calculation of the activity.
.I also need the molar extinction coefficient for Cyt c, since I have got 2/3 different values from different web sites.
.Another confusion which I have is the units for expression of the activity. Is it "Moles per min per mg protein"
.Also, do we consider the path length during the calculation?
