Dear all, I will be expressing a soluble protein in S. cerevisiae followed by cell break and Western blot of the lysate. I will break the cell using Y-PER Plus Yeast Protein Extraction Reagent. It says this method is better than the glass bead method, but my concern is will the detergent in it play havoc my protein. I know answer to this question is to check it out, but since I have some time left before I do that, I would put some thinking in this.

Has anyone had problems of detergent disrupting the protein using Y-PER Plus Yeast Protein Extraction Reagent? If yes, then I may order some glass beads.

Thank you in advance.

Best Regards,
