I am new in Y2H and doing the yeast transformation(Y2H gold strain from clontech) with my bait- pGBKT7.I am running the control all along.But the problem is my bait does not grow in SD/-trp DO plate.I am exactly following the yeastmaker yeast transformation system manual from clontech.Control is working well.So I am trying to resolve this issue.Please suggest me probable causes and basic tricks if any.Thank you.

I'm also using the clonetech system with pGBKT7 and pGAD vectors. For transformation, the GEITZ lab short protocol is working well with me, give it a try, its very quick but you gotta change it according to your own requirements.
