Hi,

I have encounter a lots of problems with the construction of yeast strain using yeast one hybrid kit from Clontech. The plasmid I have constructed refused to be digested. The plasmids involved are placZi, pHisi, pHisi-1.Have anyone face the same problem. I would like to know which lab is working with one-hybrid.
