Hey everyone:
I have some troubles in my yeast two hybrid experiments.  Becasue my bait is self-activation , even after contransformed with prey, should all  the results   turn blue  in Theoretically? But when i cotransformed  my bait and preys,  the result is not all turn blue. (the same bait vs several preys)    Why?

the prey interacts to inhibit autoactivation?

is your bait a transcription factor?

my bait is a 35AA peptide. This peptide  can be autoactivation  , either it fusioned to AD or BD.   more interestiong,  it can be autoactivation, even without AD or BD. too strange. there is no such short transcription factor  reported before..........................

my bait also had self activation. by the domain analysis, i found that my protein has one SATA domain. In general, this domain has the function as one transcription factor. could i continue to do y2h via my protein
