Hi all,

I have been designing primers for my work but I am really kind of stuck this time round.

I want to create primers with with RGC RE (response element) in it. I need to have two RGC in the primer sequence. My supervisor told me that I can create a primer with a 5bp starter, followed by a RGC RE sequence, then a 20bp spacer (random nucleotides) and followed by a RGC RE sequence before some 20+bp that anneals to the template that I have. This is so that I can create a template with two RGC in front.

The headache comes at the 20bp spacer. I have no idea what I should use as the 20bp spacer. One of my supervisors told me any random nucleotides will do. And I know nuts about using Primer3. Any help would be appreciated.

Thanks in advance!!!
