Hi, I am planning to make cell lysis buffer (10mM Tris-HCl,100mM EDTA,2% SDS) for genomic dna extraction and was wondering if I need to autoclave it after I make it? I have always been using cell lysis buffer that comes with extraction kits and it's the first time I'll be using in house buffer.

Don't autoclave SDS containing solutions, it will foam like crazy causing you to lose a lot of the buffer. Also it will denature the SDS, not that this affects its ability to act as a detergent.
You could filter it, though with the SDS, I would be surprised if anything will grow in it.
