Hi, there:

I have some problwm with my low transformation efficiency with BL21 DE3(pLys) E coli strainby chemical method. Actually, around 100-1000 times lower than common E coli such as Dh5.Any experience or suggestions?

hi, This is a common problem. Bl21 does show lower efficiency. to get transformants in fairly decent nos use larger amounts of DNA(preferably of better purification) and use freshly prepared competent cells. somehow the bl21 efficiency drops real low on storage.All the bestmaitreyi
