I am trying to isolate a plant gene by using degenerate primers designed from conserved consensus of homologous genes from other plants. I was able to get a faint PCR band of expected length (750bp) together with intense primer dimer bands. Upon reamplification, I could not get the amplicon of interest because the primer dimer tends to predominate during reamplification.
What cloning kit would you recommend for faint PCR bands? Thanks in advance!
