You must either spheroplast mycobacterial cells by adding 10 g Glycine per liter of culture at OD 3.0 and letting grow for another 24 hours or by using French Press at 14000 Psi.

Any other efficient way to disrupt Mycobacterium tuberculosis cell wall during DNA extraction?
