Hi everyone,
at the moment I'm struggling to get my plasmids (CEN plasmid with auxotrophic marker) into diploid yeast cells. In haploids its quite easy with PEG, LiAc and heatshock on cells from a fresh grown agar plate. I also tried a normally very efficient method where the yeast where first made competent and the heat shocked for a shorter time. But somehow with the diploid strais it does not work .
I would be gratefull for any suggestions or ideas how to get the cells swallowing these plasmids.
