I culture a549 (lung carcinoma) cells in DMEM with 10%FBS. Until now I used to treat my cells with benzo[a]pyrene in complete medium and then running Western Blot with the whole protein extract. The very strong (bold) bands of my control sample made me think whether it would be better to have a period of serum starvation before treatment. But I wonder which is the most effective time period (4h, 24h etc). And after that should I make the treatment in serum-free medium or not?
