This depends on the enzymes used. NEB suggests a minimum of 2 bp for EagI. I don't know about the other. If you do a sequential digest with SfiI first, you only need the 2 basepairs between the two sites.

Hello folks, I want to engineer a new restriction site, EagI, into an expression vector. but I don't have much room as there is a required SfiI site that I need to maintain. Does anyone know what the minimum distance between two resrtiction sites is for efficent digestion
