Cell Yields from Adult Rat Hippocampal Cultures

Hi,

I've just started culturing neurospheres derived from the dentate gyrus of the adult rat hippocampus. The protocol I'm using is based on the Brewer et al 2007 paper and requires me to grow my neurospheres in 24 well plates.

I don't know if anyone has experience in these type of cultures but I feel my cell yield is quite low, between 33,000-90,000 cells/ml and I have a total volume of 3ml of cells. With so few cells I've only grown the cells at 50,000 cells per well but would also like to try 100,000 cells per well, which clearly I don't have enough for.

In peoples experience are these yields exceptionally low or about normal. I would expect there to be quite a bit more cells from the adult tissue, just that the level of proliferation observed in culture would be lower than using embryonic tissue.

Any advice is greatly appreciated!!
