endotoxin amount defined as EU/ml

want to know how endotoxin amount EU/ml could be converted into µg

and how much endotoxin could be injected into a mouse w/o any physiological changes/avoid lethality??

Depending on the (bug) source of the endotoxin, the conversion from endotoxin units to weight quantities will vary and the same goes for amounts by injection that provoke response.

Are you talking about E coli endotoxin?

I am not really familiar with microbiological details like E.coli strains, etc...but sure that its E.coli

I perform maxiprep (Endotoxin Free NucleoBond Plasmid DNA Purification, Macherey-Nagel) to purifiy high amount of different plasmids. Afterward I additionally perform endotoxin removal (Detoxi-Gel Endotoxin Removing Gel, Pierce) to remove residual endotoxin, finally i performed an LAL assay to quantify remaining endotoxin of the sample.

i am a little paranoid of endotoxins, because i have to inject the sample into mice and dont want to be too harmful. in earlier times i injected my DNA without any purifications/removals, which led to very strange looking livers - i thought that this was the effect of endotoxins??

For E. coli, 1 ng is a threshold dose for man and rabbits - don't have data for mice.

1 EU is about 0.2 ng

Normalised the FDA suggest that 10EU =1 ng of Endotoxin.

can you give a ref? I'm not familair with "normalized" LPS.
