I am looking to develop a microplate assay using fluorescein conjugated antibody as the detection agent. I haven't run this sort of assay before and can't find a protocol. I am assuming I can run it like a normal ELISA, using a fluorescence plate reader. Once the conjugate has been added to the plate, does it need any form of protection (e.g. like an anti-fading agent as in IFA)? Are there any other specific considerations I need to be aware of, either in the protocol or the hardware? Any advice or links would be much appreciated.
