Hi, does anyone know the kind of strategy to perform mutagenesis on large plasmid of around 30kb (20+kb insert) or on articifial chromosomes?
I know for large plasmids there is a XL mutagenesis kit for plasmid >8kb from strategene, but i'm not sure if it will work plasmids up to 30kb.  If anyone can direct to any resources it will be great. Thanks in advance.

I would subclone a smaller fragment into a regular vector, do the mutagenesis, then subclone it back in.
