Hi folks,
I have been doing real time PCR from the last couple of years, but is stuck up with an issue- I did expression (Taqman) assay for microRNA and presented data as fold change using delta-delta Ct method using standard internal reference as miR48. However one of reviewer of our paper want the data to be presented as delta Ct. What is the best way to to represent data in delta Ct. To me it appear that it will complicate figure and will not be easy to interpret? Any suggestion for best presentation or published paper will be great.
My second question is how can I calculate copy number in microRNA taqman assay?

Thanks for your input.

This doesn't exactly answer your question I think, but I hope you can find it useful:

Clinical Chemistry 55:4 611–622 (2009)
Special Report:
The MIQE Guidelines: Minimum Information for Publication of Quantitative Real-Time PCR Experiments

http://www.clinchem....eprint/55/4/611

SUMMARY: Following these guidelines will encourage
better experimental practice, allowing more reliable
and unequivocal interpretation of qPCR results.
