I am running homogenized human tumor samples and am consistently having problems with one of my groups. There are 6 treatment groups and when I coomassie stain the membrane 5 of the 6 always look fine and evenly loaded and the final treatment ALWAYS has almost no protein showing. I am confident I am loading the same amount of protein into each well (I have tried 2 different methods of protein determination and have gotten the same results).
I don't know whether something in these tumor samples is blocking them from running. Does anyone have any suggestions for what I can try?
