What is the best way to evaluate the in vitro toxicity of a drug?

I would try to assess the % viability of different unstimulated ( or better if stiumulated?) cell lines with a simple flow cytometric live/dead staining , but expecially hepatocytes and lymphocytes after exposure for X ( how many?) hours to the test drug at different concentration.

Do you have any suggestions?
