Hi everyone,

does anyone know how to enhance the last homologous recombination step in bacteria? I´ve the worst recombination rate ever. I even didn´t use any dilution steps after the overnight incubation before i platted the cells on the selective media (Chloramphenicol, because i used a marker replacement strategy and saccharose to get rid of the plasmid).
Any tipps like rising/lowering incubation temperature?
