I am planning to make my first luciferase reporter construct using the promoter region of my chosen gene. What would be the best source to PCR that region for cloning? If I need the promoter to be of human origin, can I just take a human cell line, isolate DNA and PCR it? Any tips for DNA isolation and PCR in this case?

Hi,
I did something similar.
I bought human genomic DNA from Biochain, but I think also extraction from cell line should work. Qiagen for sure has a kit for the DNA extraction.
