Hi,
I did some confocal imaging few months ago and sent the result to the journal. The reviewer asked me to put X-Y plane to show its 3D. I really dont know how to do it? Can any one help me? Our imaging system is leica SP5.

I assume you mean an XYZ image? Did you take a stack of images of your specimen? For example 10-20 sections through one field of view? If you did then the software should allow you to display these images as an overlayed 3D stack. And then should give you the option to view the z plane. I'm not familiar with Leica but it is very straight forward on the Olympus confocal.
