I tried to boil samples 15 minute at 100 C and than i used in my PCR but i did not any results of my PCR product. I used cells from buccal swabs. I know that is some a commercial kits for direct PCR but I want to try this method without this kits and I want to request anybody for any simple guidance. Thank you very much for your help.

Try to boil samples 5 minutes in 0.2M NaOH.

This technique is reported to be useful on plants and their cell walls are much stronger than human so I guess it may working.
