Hi!

I'm very confused this morning with my results! I did a transformation in Methylobacterium extorquens to produce knockout. I did check my plates this morning and WOW, I got few colonies... but they're growing on the petri cover, not on the agar?!?! I use a minimal medium with methanol and I sealed the plates.

Do you have any ideas about what happened? Of course, my plate was incubated in the good position.

What do you mean with "good position"??? Upside down?

Did you get any medium on the lid of the petridish when pouring them? Or did you have any excess water in your plate so your colonies were washed to the cover?

Yes, upside down.

No, I didn't have any medium on the lid and my plates were totally dried before I incubated them.
Do you think it's possible that the colonies aren't well attached to the agar, and fall when the size became bigger? That's the only "logic" explanation I can figured!

I have never heard of falling bacterial colonies....but my experience with bacteria is not too big.

Are your bacteria quite mucous????

Not mucuous, in fact when I try to repick a colony it's like mercury, you know, the total colony move when I touch it.

Not familiar with jumping colonies but sound slike fuin. Try incubating the plate uninverted.

Are you sure what you see are actually colonies by the way?
