Hello everyone!

I am new to the Real-Time PCR field and I would like to ask some questions:

1. First, we buy the ''home made '' SYBR Green (it is not a kit, we just dilute it), but we always have to measure it and adjusting it before the QPCR. Why do we have to do that everytime?

2.We measure the Sybr Green in 495nm in nanodrop and the indication should be 0.4 mean?Why do we use this wavelength and what does this o.4 indicate?
