Preparation of tree tissue for western blot

Hi!!

I would like to know if somebody knows which is the best way to prepare samples of trees such as cherry or peach tree for western blot. I have usually made this for Nicotiana, but in the case of trees what I get when I mix the sample (leaves ground to powder under liquid nitrogen) with the buffer containing urea, b-mercaptoethanol, etc, is a sticky supernantant that runs horrible in the gel.

I would appreciate very much if somebody could give me a better recipe for the rupture buffer, of any tips about the preparation of the samples.
