Thawing and Plating

Hi,
do I have to plate thawed lymphocytes before extracting RNA or performing a flow cytometry analysis? If yes, Do I have to use medium with or without serum?

No on the RNA extraction. Yes on latter only if by culturing and allowing several passages will increase the expression of marker you are wanting to evaluate by flow cytometry. If the cells have plenty of marker of interest then plating would not be necessary. Make sure you perform proper thawing procedure and adequately remove DMSO. Hope this helps.
