In my M.Sc. thesis work I  found some E. coli with Ciprofloxacin resistance carrying  plasmids of less than 6kb. After plasmid curing they became sensitive to ciprofloxacin.  When I transformed DH5α with these plasmids they became resistant to cipofloxacin but I couldn't find any plasmid after plasmid extraction process. I carried out both calcium chloride method and electroporation techniques for transformation. For extraction process I used both Doly methad and commercial kit.

Please somebody give me some light.
