Hi, Why do you remove the agar? If you are trying to get the virus for recovery you can just stab through the agar with a pipette tip.

Hello, it seems like you know a thing or two about cell culture.
I have a question about a plaque assay I am trying to do. I know the virus (MCMV) I am using works; however, the cells (3T3, grown in DMEM+10%FCS+PSG+NaHCO3) keep coming off the plate (I believe when I remove the agar). Do you have any suggestions?

Hi Swizla21, I haven't worked on THP-1 cells before, although a lot of the principles of general cell culture will apply. All I know of them is that they are a cell line that can differentiate into macrophages on stimulation, which is usually done with LPS (lipopolysaccharide).

Many thanks for your words of wisdom regarding protein tags. Do you happen to know of any examples where the addition of a tag ultimately altered protein function through mis-folding?

I found in forum archives a comment by you in which you suggested that one way to distinguish on cell line from another is to use cell-specific primers and PCR. I need to do just that, and I have been searching online for a few hours but have not been able to find a protocol for such a procedure.
