I was wondering: wich tests would you suggest if you have a set of compounds that you want to screen for cytotoxic activity on some cancer cell lines

You could start with the trypan blue exclusion assay to see if there's any change in cell growth rates, its cheap and easy but not good for high throughput.

Apoptosis assays include annexin V binding, caspase activity, BrdU incorporation, DNA laddering, and many more.....

You could also look for senescence using beta-galactosidase activity.

so the tests that I know and that can be useful in this setting are:
Cell Viability assays ( like Trypan Blue, or 7AAD or PI)
Cell Proliferation assays ( like the CFSE assay)
Cell Cycle Analysis tests ( like Rnase/PI in fixed cells)
Apoptosis assays ( like the Annexin V assay)
Senescence assays

do you know something equivalent, or at least comparable, to the MTT assay, that I can perform in flow cytometry?
