We're looking to transiently transfect gastric epithelial cells with a certain gene but have obtained low efficiency with the promega tfx system. We're now looking for a virus vector system that might offer better efficiency. Can anyone recomend a good (fast, easy, cheap) system?
Thanx!

hi
you can use the one mentionned in this topic (pBABE) :
http://www.protocol-online.org/forums/inde...?showtopic=5152or you can use pLNCX vector, or pSUPER-retro...

I would definitely recommend using a lentiviral system over a retroviral system when dealing with primary cells.
You can also get pretty decent transfections in primary cells (I tried with fibroblasts) with JetPEI transfection reagent.

I agree with warsel, lenti is the way to go if you have the facilities to make them, especially if your cells don't divide.

Viral vectors are a tool commonly used by molecular biologists to deliver genetic material into cells.

Following is the best lentiviral vector(pCSCG) that I have ever used. Gives very good efficiency even in primary epithelial cells. Only disadvantage(?)is it has GFP but no drug election.
