HI

Is it true that by adding a few nucleotides to the restriction sites of some endonucleases you will increase its efficiency?
I think I heard that in a lecture, but my Pi have never heard of it...so, I am confused now.... sad.gif

You add the extra bases to the end ofteh RE site ,to give hte enzyme something to hold onto. Check out the NEb website and see how many extra bases are required to improve your Re's efficiency.
