My wish

Posted by Adrian K, 09 February 2011 · 2,811 views

Finally I got myself a new workstation for me to work on my papers and thesis, and of course I burnt all my savings which is around ~USD800.Just finished the first draft of my paper today, sent to both of my boss and hope they should stop angry with me. I still owe them another paper, since the patent of the primers was done and I should be able to write them the paper. I had previously done some other work and had wrote a short communication, but my boss think it will be good if I can do more work and publish as a full article. Now had to do some re-planning.Molecular modelling of a hypothetical gene is a heavy task for me, I never had formal lesson in structural biology or related course. Docking sounds greek to me as well. I am good in repairing PC but that doesn't makes me a bioinformatic geek. I had just attended a training in using Discovery studio and my boss expect me to produce a paper using the knowledge i gained from the workshop. There are 3 licenses in other department which they are a bit reluctant for us to use it. It is not like we can install the software in our computer and run it remotely: we have to connect to a server to get our work done. Geez life is difficult. Now that I had cloned 10 genes and have to express it. I never knew that commercial sequencing company were unable to sequence highly GC rich product. There goes my 2 genes which is ~3kb long... I got no idea yet on how to tackle the problem. Maybe the conventional sequencing method by interpreting the long PAGE gel?So, I told my boss I had cloned 10 genes and is on to way to do expression of the recombinant protein. She told me that I should be able to publish 2 papers from there, but I do not agree with her. For the all 10 to be successfully expressed is one huge challenging job, but the thing is we do not have a proper protocol yet to assay our recombinant protein, this is indeed a big trouble which yet to be solved. Any lectins specialist here?At the end of this month, I have to do my candidature defense. I have not prepare my slides yet as I have yet to have some preliminary results. For this brand new year, I wish I could finish my project, submit at least 3 papers and get a job as I really need money badly. But then back to reality, my stipend only supports me till this April, 2 months more to go.I really pray for luck to come this time. And yes, Zaraki Kenpachi is my favorite BLEACH character. This is one of the great song I enjoy very muchFrom http://www.azlyrics....obefree.html"(I Wish I Knew How It Would Feel To Be) Free"I wish I knew how it would feel to be freeI wish I could break all the chains holding meI wish I could say all the things that I should say Say 'em loud say 'em clearFor the whole wide world to hear I wish I could shareAll the love that's in my heart Remove all the bars that keep us apartAnd I wish you could know how it is to be meThen you'd see and agree that every man should be free I wish I could be like a bird in the skyHow sweet it would be if I found I could flyWell I'd soar to the sun and look down at the sea And I'd sing cos I know how it feels to be freeI wish I knew how it would feel to be freeI wish I could break all the chains holding meAnd I wish I could say all the things that I wanna say Say 'em loud say 'em clearFor the whole wide world to hear Say 'em loud say 'em clearFor the whole wide world to hear Say 'em loud say 'em clearFor the whole wide world to hear One love one bloodOne life you've got to do what you should One life with each otherSisters, brothersOne love but we're not the sameWe got to carry each other Carry each otherOne One One One One...I knew how it would feel to be freeI knew how it would feel to be free
